摘要
目的:测定杜仲叶中绿原酸的含量及探究生物合成机理;方法:用30%甲醇回流20min,反相液相色谱法测定杜仲叶中绿原酸含量;色谱柱为Kromasil C18柱,流动相为甲醇-0.02%磷酸溶液(22:78),波长327nm,流速1.0ml/min.绿原酸在9.78-244.5μm/l呈良好线性关系,r为0.9998,回收率为97.9%:结果:通过对10年树龄的光皮杜仲树叶4~11月绿原酸含量的分析,叶中含量平均为2.55%,6月份达到最高;结论:杜仲叶的绿原酸含量与体内的生长激素调节有关.
Aim to Determination and discuss biosynthesis of Chlorogenic Acid in Leaf of Eucommia ulmoides Olive Method the conditions for the extracting chlorogenic acid in the Eucommia Ulmoides Olive leaves were optimized as follows: by 30% methanol for 20 minutes; determined by RP - HPLC. The chlorogenic acid was separated on kroma- sil C18 column, with CH3OH-0. 02% H3PO4 (22: 78) at a flow -rate of 1.0ml/min and detected at UV 327nm. The calibration curve was linear in the range of 9.78 - 244. 5 μg/ml ( r = 0. 9998 ) . Results the analysis of the content of chlorogenic acid in the leaf of smooth leather Cortex with 10 years old was earned out and the result indicated the content of chlorogenic acid differed greatly in the leaf; the content of chlorogenic acid in leaf was higher on June. Conclusion biosynthesis of chlorogenic acid in leaf of Eucommia ulmoides is controned by growth hormone.
