摘要
作者用黄疸出血群赖型钩体外膜免疫BALB/c小鼠,取脾细胞与SP 2/0骨髓癌细胞融合,筛选出三株抗问号状钩体外膜 McAb。显凝试验(MAT)表明 McAb的E4B11C9株与选用的黄疸出血群的13型钩体(占选用的100%)均发生凝集反应,而选用的其他18群钩体(占选用的100%)和非致病性Patoc Ⅰ株钩体,伊利尼细螺旋体均为MAT阴性,MAT效价低于1:25。从而证明 E4B11C9 McAb具有黄疸出血群钩体的群特异性。
Three McAb were produced against anouter envelope preparationl from Leptospira.interrogans, serovar Lai by fusion of SP2/0myeloma cells with immune BALB/c micespleen cells. The fusion rate was 96% andthe antibody positive rate was 50%.One ofthe hybridomas, E_4B_(11)C_9, reacted with 13of tho 13 serovars of the Icterohaemorrha-giae serogroup in microscopic agglutinationtest (MAT) but did not react with the 18representative serovars of L. interrogansand L. biflexa serovar patoc and Leptone-ma illini.For all non-reactive serovars theMAT titres were greater than 1:25. TheMcAb, E_4B_7G_5, reacted similarly with allserovars except smithi and tonkini. E_4B_7D_4reacted also similarly with all serovarsexcept serovars birkini, ndambari,bogvere,smithi and tonkini. Therefore, 3 McAb showed serogroupspecificity and partial serogroup specificityby agglutination. The agglutination titreswere high and hybridomas were stable, soit might be useful in providing a simple,rapid method for the classification andidentification of clinical isolates such aspathogenic L. interrogans in place of thecomplicated and time-consuming conven-tional methods.
