猪圆环病毒2型河北分离株的基因型鉴定及其毒力分析 被引量:8

Genotype identification and virulence analysis of porcine circovirus type 2 isolates from Hebei province

摘要 为了明确河北部分地区猪圆环病毒2型(PCV2)流行毒株的基因型及其毒力,应用免疫过氧化物酶单层实验(IP-MA)对从保定、唐山和衡水分离的3株PCV2进行了鉴定,分别命名为PCV2-BD1A、TSh1和HSh1。以PCV2 Cap基因为靶基因,运用PCR-RFLP技术对3个分离株进行了基因型鉴定,结果分离株BD1A和TSh1为2H基因型,分离株HSh1为2I基因型,提示2H基因型仍然是当前河北地区PCV2流行毒株的优势基因型。通过与已知PCV2强毒株(PCV2-40895)Cap蛋白的氨基酸序列比较分析,推测PCV2-BD1A、TSh1和HSh1 3个分离株均为强毒力PCV2。 In order to clarify genotype and virulence of porcine circovirus type 2(PCV2) prevalence in some regions of Hebei Province, three porcine circovirus type 2(PCV2) isolates named PCV2-BD1A, TSh1 and HSh1 from Baoding, Tangshan and Hengshui were identified by immunoperoxidase monolayer assay(IPMA). Two genotypes including 2H(BD1A and TSh1 isolates) and 21(HSh1 isolate) were identified according to PCV2 Cap gene via PCR and restriction fragment length polymorphism( PCR-RFLP ) analysis, which indicated 2H was still the dominant genotype among PCV2 isolates recently prevalent in Hebei Province. Based on the comparison of their amino acid sequences of Cap protein with that of proved high pathogenic strain PCV2-40895, it was speculated that PCV2-BD1A, TSh1 and HSh1 were high virulent isolates.
