Expression of hyperplasia suppressor gene in breast cancer tissues and its significance

摘要 目的:探讨新的增殖抑制基因(hyperplasia suppressor gene,HSG)在乳腺癌组织中的表达及其意义。方法:应用免疫组织化学和原位杂交法检测55例乳腺癌标本和18例癌旁正常乳腺组织标本中HSG的表达。应用Image Prop lus数码图像分析系统测定组织切片的积分吸光度(D)值,分析其与临床病理参数的相关性。结果:HSG mRNA及其蛋白的阳性染色主要集中于细胞质,呈棕黄色颗粒。55例乳腺癌标本中HSG mRNA及其蛋白的阳性表达率分别为40.00%(22/55)和41.82%(23/55),而癌旁正常乳腺组织的表达率分别为83.33%(15/18)和88.89%(16/18),2组比较,差异有统计学意义(P<0.05)。在乳腺癌组织中HSG mRNA及其蛋白的低表达与组织学分级、淋巴结转移和雌激素受体(estrogen receptor,ER)有关(P<0.05),与肿瘤大小和年龄等无关(P>0.05)。结论:HSG在不同乳腺组织中的表达量不同,提示HSG可能与乳腺癌的发生、发展有密切关系。 Objective:To investigate the expression of hyperplasia suppressor gene (HSG) in the tissues of breast cancer and its significance. Methods: Immunohistochemistry method and in situ hybridization technique were used to detect the expression of HSG in 55 breast cancer samples and 18 normal adjacent breast tissues, hnage Pro plus image-processing anaiysis software was used to detect the integral optic density(D) values of the slides of breast tissues, and the correlation of D value with clinicopathologic features was analyzed. Results:The positive staining of HSG mRNA and protein were demonstrated as pale brown particles which mainly accumulated in the plasma of cells. In 55 cases of breast cancer tissues, the positive rate of HSG mRNA and protein was 40.00% (22/55) and 41.82% (23/55), respectively. In the normal breast tissues, the positive expression rates of HSG mRNA and protein were 83.33% (15/18) and 88.89% (16/18), respectively. The difference was significant between the two groups (P 〈 0.05 ). In breast cancer tissues, the weak expressions of HSG mRNA and protein were associated with histological grade and estrogen receptor (ER) ( P 〈 0.05 ), but were not associated with the tumor size and age( P 〉 0.05 ). Conclusion :The expression rates of HSG in different breast tissues are at various levels. It indicates that there is a close relationship between HSG and the occurrence and development of breast cancer.
