摘要
本文采用压缩空气吹展DNA的制样方法,使得DNA链在云母表面大致向着同一方向充分舒展,为DNA线度的AFM测量打下了基础。对λDNAHindⅢ依次进行了酸变性、复性、DNaseⅠ降解处理,并分离一种抗内切酶降解的组分。AFM研究表明该组分可能是一种新的多链DNA。
Abstract In this paper,DNA sample for AFM imaging is prepared by blowing the solution with compressed air to fully stretch the DNA strands on the surface of mica,which lay a foundation for measuring the dimensions of DNA.λ DNA Hind Ⅲ is treated sequentially with acid denaturation,renaturation and DNase I digestion,and one component resisting digestion has been separated from the mixture.AFM studies reveal that this component may be a new kind of multi stranded DNA.
