摘要
目的用不同方法观察家蝇三龄幼虫血细胞的分类、各类血细胞计数及感染大肠杆菌后血细胞的形态变化。方法(1)应用姬氏染色结合相差显微镜对家蝇三龄幼虫血细胞形态进行观察并分类。(2)倒置显微镜下观察家蝇三龄幼虫血细胞总数(THC),姬氏染色法计数各类血细胞(DHC)。(3)观察大肠杆菌感染后家蝇幼虫血细胞形态的变化。结果(1)家蝇三龄幼虫血细胞可分为原血胞、浆血胞、粒血胞、珠血胞、类绛血胞5类。其中浆血胞又分为大核浆血胞和小核浆血胞两种。(2)家蝇三龄幼虫血细胞总数为7090±651/μl,其中原血胞约1.2%、浆血胞约50.4%、粒血胞11.4%、珠血胞约36.2%、类绛血胞约0.8%。(3)感染大肠杆菌后,浆血胞发生空泡、细胞变形等变化。结论家蝇三龄幼虫血细胞分为5类,血细胞总数为7090±651/μl,其中浆血胞是主干血细胞,并参与感染后的细胞免疫。
Objective To observe the hemocytes classification , and the number of the different bemocytes of the 3rd stage larvae of Musca domestica by different methods, and to research the changes of hemocytes morphous after being infected by Escherichia coll. Methods ( 1 ) The hemocytes morphous and classification of the 3rd stage larva of Muaea domestica were observed by both Giemsa staining and phase contrast microscope. (2) The number of total hemocytes (THC) in the hemlymph of the 3rd stage larva of Musea domestica were counted under inverted microscope, and the number of the different hemoeytes (DHC) were counted with Giemsa staining. (3) The changes of the hemoeytes morphous of the Musea domestiea larvae after being infected by E. coli were observed. Results ( 1 ) The hemoeytes of the 3rd stage larva of Musca domestiea was divided into prohemocyte, plasmatoeyte, granuloeyte, sphruloeyte and cenoeytoid. The plasmatocytes was divided into big nucleus hematocyte and mieronueleus hematocyte. (2) The number of total hemoeytes (THC) of the 3rd stage larva of Musca domestica were 7090 ±651/μl, and the percentage of prohemoeyte, plasmatoeyte, gTanuloeyte, sphruloeyte and cenceytoid were about 1.2%, 50. 4%, 11.4%, 36. 2% and 0. 8%. (3) The vacoolation and morpholysis were found in plasmatoeytes after being infected by E. coil Conclusion The hemoeytes of the 3rd stage larvae of Musea domestiea was divided into five types. The plasmatocytes were dominant, and they participated in cell immunity.
