当归提取物对内皮细胞迁移能力及明胶酶表达的影响 被引量:2

摘要 目的研究当归提取物对内皮细胞迁移能力及明胶酶表达的影响。方法应用迁移实验研究了当归提取物对内皮细胞迁移能力的影响,应用半定量RT-PCR技术及免疫印迹杂交技术分别在mRNA及蛋白水平检测了当归提取物对明胶酶表达的影响,同时应用明胶酶谱法观测了当归提取物对明胶酶活性的影响。结果0.1、1.0及10.0g/L浓度的当归提取物可明显抑制佛波酯诱导的内皮细胞迁移,抑制率分别为(30.44±7.01)%、(52.30±7.48)%及(74.77±3.15)%;同时当归提取物可影响明胶酶B的mRNA及蛋白的表达,并可下调其活性,10.0g/L浓度的当归提取物对明胶酶BmRNA、蛋白及酶活性的抑制率分别为(33.75±7.79)%、(72.36±4.02)%及(44.52±11.75)%。当归提取物可明显抑制明胶酶A蛋白表达及活性,10.0g/L浓度时抑制率为(72.34±5.07)%、(53.70±6.46)%,但对明胶酶AmRNA的表达无明显影响。结论当归提取物可在体外抑制佛波酯诱导的内皮细胞迁移,此作用可能是通过其对明胶酶表达或活性的抑制来实现。 Objective To investigate the effect of angelica extract on the migration of endothelial ceils and the expression of gelatinases. Methods In vitro migration was used to determine the effect of angelica extract on the migration of endothelial elicited by phorbol 12-myristate-13-acetate (PMA). Semi-quantitative reverse chain reaction and western blot were used to detect the effect of angelica extract on the expression of gelatinases at messenger ribonucleic acid (mRNA) and protein levels respectively, while the proteolytic activities of gelatinases were assessed by zymography. Results The migration of endothelial cells elicited by PMA reduced to(30.4±7.01)%, (52.30±7.48)% and (74.77±3.15)% respectively when incubated with 0.1, 1.0 and 10.0 g/L angelica extract, the drug also can reduce the expression and activities of gelationase B. At 10.0 g/L concentration, the inhibition ratio of gelatinase B mRNA,protein and activities are (33.75±7.79)%, (72.36±4.02)% and (44.52±11.75)% respectively. The protein expression and activities of gelatinase A can be inhibited by angelica,the inhibition ratio were (72.34±5.07)% and (53.70±6.46)% respectively at a concentration of 10.0 g/L, while the expression levels of gelatinase A mRNA was not affected. Conclusion The expression and activities of gelatinases were downregulated in endothelial cells when incubated with angelica extracts, this is the possible mechanism that angelica extract inhibit the migration of endothelial cells elicited by PMA in vitro.
