摘要
目的探讨蝎龙液浸膏对动物体内外抗凝血、纤维蛋白溶解及抑制血栓形成的作用。方法对蝎龙液浸膏进行活化部分凝血活酶时间(APTT)、凝血酶原时间(PT)、凝血酶时间(TT)及复钙时间测定,采用纤维蛋白平板法测定蝎龙液浸膏纤溶活性。用Chandler法制备家兔体外血栓和半体内血栓模型;用Reyers法制备大鼠静脉血栓模型。测定蝎龙液浸膏对家兔体外血栓;半体内血栓;大白鼠静脉血栓的抗栓效应。结果蝎龙液浸膏能轻度延长家兔血浆APTT、PT、TT及血浆复钙时间,在普通纤维蛋白平板上能溶解纤维蛋白,对家兔体外血栓;半体内血栓;大白鼠静脉血栓干湿重均有明显减轻作用,与生理盐水对照组比较,差异均有统计学意义(P<0.05或P<0.01)。结论蝎龙液浸膏具有抗凝作用,能间接溶解纤维蛋白、抑制动物血栓的形成。
Objective To study anticoagulation and antitrombotic effects of Xielongyeextract in vitro and vivo. Methods APTT, PT, TT and PRT were determined by kit illustration and fibrinolytic activity of Xielongye was determined by fibrin plate. By means of Reyers's venous thrombosis model in rats, the Xielongye was given by mouth at 1 mL·kg^-1. NS was used as negative control and thrombolytic effects was observed and Chandler's mixed thrombosis model in vitro. Results The experiment in vitro and in vivo showed that Xielongye could prolong slightly APTT,PT ,TT and PRT( P 〈0.01) ,and it has acting on fibrin in the fibrin plate, the dry-wet- weights of venous thrombus were marketly lightened. Comparison with control group of NS, the difference has statistical significance (P 〈 0. 05, P 〈 0. 01 ). Conclusion Xielongye has obviously anticoagulation and thrombolytic effect in vitro and vivo .
