胃癌细胞C-myc蛋白对他莫昔芬的应答

摘要 目的:观察C-myc蛋白在胃癌细胞系BGC823中的表达情况,探讨胃癌细胞BGC823经他莫昔芬(TAM)处理后,C-myc蛋白表达的变化.方法:使用10 5MTAM处理胃癌BGC823细胞系24 h后,通过免疫荧光细胞化学的方法及细胞计数,检测C-myc蛋白的表达变化,并通过WST1法检测细胞活力的变化.结果:C-myc在胃癌BGC823中高表达,经10 5 M TAM处理24 h后BGC823细胞活力明显下降(P<0.05,)同时C-myc的表达减弱(P<0.05).结论:C-myc在胃癌细胞BGC823中发挥生物学作用且与TAM介导的BGC823细胞活力下降相关. Objective: To observe the expression of C-myc in gastric cancer BGC823 cell and research the alteration of C-myc expression in gastric cancer BGC823 cell after the treatment of Tamoxifen. Methods: By fluorescence immunocytochemistry staining and cell counting of BGC823 cell treated with 10^-5M Tamoxifen for 24 h, detected the expression alteration of C-myc, at the same time, cell energy was detected by WST-1 method. Results: The over expression of C-myc was detected in gastric cancer BGC823 cell. The decrease of expression of C-myc (P 〈 0.05) and cell energy (P 〈 0.05) were observed in gastric cancer BGC823 cell treated with 10^-5M Tamoxifen for 24 h. Conclusion: C-myc plays an important biological role in the gastric cancer BGC823 cell and is related to cell energy change mediated by Tamoxifen.
