毁损海洛因依赖大鼠伏隔核对戒断症状及血浆β-内啡肽的影响 被引量:3

摘要 目的观察毁损海洛因依赖大鼠伏隔核(NAc)对戒断症状及血浆β-内啡肽(β-EP)的影响。方法①海洛因依赖组(实验组)按逐日递增原则皮下注射海洛因建立海洛因依赖模型,首日每次剂量3mg/kg,雌雄不拘,每日2次,连续9天皮下注射海洛因;对照组注射等量生理盐水。②毁损NAc:毁损电极插入NAc,通直流电进行毁损。实验组大鼠20只,分为实验Ⅰ组(海洛因依赖组)和实验Ⅱ组(依赖后行NAc毁损),每组10只;对照组大鼠20只分为对照Ⅰ组(NAc未毁损组),对照Ⅱ组(NAc毁损组),每组10只。比较实验Ⅰ组和实验Ⅱ组戒断症状的变化;③β-EP检测:实验Ⅰ组、实验Ⅱ组、对照Ⅰ组、对照Ⅱ组大鼠取躯干血置入抗凝试管中,混匀离心,放射免疫法测定4组大鼠血浆β-EP水平,比较各组大鼠血浆β-EP水平的变化。结果①戒断症状:毁损大鼠NAc后,除湿抖症状外,其他戒断症状均较毁损前减轻(P<0.01);②血浆β-EP:与对照Ⅰ组比较,实验Ⅰ组血浆β-EP水平下降,对照Ⅱ组血浆β-EP水平上升(P均<0.05);实验Ⅱ组血浆β-EP水平上升,与实验Ⅰ组比较差异有显著性(P<0.05)。结论毁损NAc后,海洛因依赖大鼠的戒断症状减轻;NAc参与β-EP水平的调节。 Objective To observe the changes in withdrawal signs and plasma β-EP after ablating the NAc in heroin-dependent rats.Methods SD rats were divided into four groups of 10.TestⅠ and testⅡ groups were made addicts by starting with heroin 3 mg/kg hypo bid and increasing the dosage every day for a total of 9 days,while the controlⅠ and Ⅱ groups were given equivalent volumes of NS.Electric ablation of the NAc nucleus was performed in testⅡ and controlⅡ groups.The withdrawal signs were compared between the two test groups following the naloxone therapy.Finally,blood samples were taken to determine the level of β-EP for comparison between the four groups.Results The withdrawal signs were less marked in testⅡ group than in testⅠgroup(after NAc ablation,P〈0.05).The level of β-EP in blood plasma was found lower in testⅠ group than in controlⅠ group(after addiction,P〈0.05),and higher in controlⅡ and testⅡ grouops(after NAc ablation,P〈0.05).Conclusion After the NAc is destroyed,the withdrawal signs are alleviated.The NAc participates in the regulation of β-EP.
