摘要
通过对五个批次的无特定病原体(SPF)鸡胚进行鸡新城疫病毒(NDV)、鸡传染性支气管炎病毒(IBV)同胚接种,收获72—120h死胚及120h活胚尿囊液,分别测定NDV和IBV的病毒含量,结果NDV病毒含量达到了10^8.3-10^8.7EID50/0.1mL,IBV的病毒含量达到了10^6.3-10^6.5EID50/0.1mL。两种病毒含量均达到《中华人民共和国兽用生物制品规程》(2000年版)中鸡新城疫、传染性支气管炎二联活疫苗病毒含量要求的标准。表明该方法即可降低成本,又简化了生产工艺。
Newcastle disease virus and infectious bronchitis virus were inoculated in the same SPF embryo for five groups. The allantoic fluid of the embryos which died in 72 - 120 hours or lived in 120 hours were harvested and then the embryo infectious dose (EID50) of NDV and IBV were detected respectively. The results indicated that the EID50 of NDV was 10^8.3 - 10^8.7/0. 1mL and the EIDso of IBV was 10^6.3 - 10^6.5/0. 1mL. The virus concentration of five groups allantoic fluids reached the standard of newcastle disease and infectious bronchitis vaccine (living vaccine) which were required in the manufacturing and examination regulations of the People' s Republic of China used in animal vaccine (2000 versions). This test showed that the two kinds of virus cultured in same SPF embryo could reduce cost of vaccine manufacture and simplify technology of vaccine manufacture.
