摘要
目的观察B组柯萨奇病毒感染对大鼠心肌来源的H9c2细胞microRNA表达谱的影响。方法用B组柯萨奇病毒3型Nancy株(CVB3)攻击成层和未成层的H9c2细胞,6h后收获总RNA,基因芯片检测microRNA表达谱。结果比对未成层与成层的H9c2细胞的microRNA表达谱,H9c2细胞成层后有54种microRNA表达显著下调,7种microRNA表达显著上调。与未感染病毒的未成层细胞比较,CVB3感染的未成层H9c2细胞有35种microRNA表达显著下调,9种microRNA表达显著上调。与成层H9c2细胞比较,CVB3感染成层H9c2细胞后,有20种microRNA的表达显著下调,16种microRNA的表达显著上调。结论细胞生长状态、CVB3攻击均显著改变了H9c2细胞microRNA表达谱。
Objective To observe the microRNA expression differentiation of rat myocardium-derived cell H9c2 infected with coxsackievirus B. Methods Confluent and unconfluent H9c2 cells were attacked with coxsackievirus B3 Nancy strain ( CVB3 ). Six hour later the total RNAs were harvested and were applied to the microRNA expression profiling analysis by microarray. Results The expressions of 54 microRNAs were downregulated and 7 were up-regulated in confluent H9c2 cells compared with unconfluent H9c2 cells. The expressions of 35 microRNAs were down-regulated and 9 were up-regulated among the unconfluent H9c2 infected with CVB3 compared with unconfluent H9c2 cells without CVB3 infection. There were 20 microRNAs down-regulated and 16 up-regulated in confluent H9c2 infected with CVB3 compared with confluent H9c2 cells without CVB3 infection. Conclusion There are differentiations of microRNA expression profiles of H9c2 at different cellular status and with coxsackievirus infection.
