摘要
目的探讨热化疗联合对宫颈癌细胞耐药的影响。方法构建survivin基因的干扰RNA载体,将其转导入Hela细胞,并应用Western blot进行鉴定。按临床常用剂量,用0.2mg/L、0.5mg/L、1.0mg/L、2.0mg/L、3.0mg/L、4.0mg/L顺铂(DDP)处理Hela细胞,并联合43℃加热,用噻唑蓝比色法(MTT法)检测Hela细胞在不同条件下的生长。结果两种细胞在不同浓度的顺铂作用下,未转染细胞增殖率高于转染细胞;43℃加热,两种细胞在不同浓度的顺铂作用下比较,未转染细胞增殖率低于转染细胞。结论热化疗联合作用可以逆转人宫颈癌细胞的耐药,为进一步研究预防人宫颈癌耐药的发生奠定基础。
Objective To study the effect of thermotherapy with chemotherapy on cervical cancer cell growth. Methods A eukaryotic expression vector carrying human anti-survivin gene was constructed and transfected into Hela cells and then MTT assay was used to measure its growth at different conditions. Results The increment rate of cells without survivin was lower than that of cells with survivin, but under the heating, the increment rate of cells without survivin was higher than that of cells with survivin. Conclusion Thermotherapy with chemotherapy can reverse the drug-endurance of cervical cancer cells, which may be helpful to research on its prevention of cervical cancer.
