血红蛋白对琼脂糖凝胶尿蛋白电泳分析的影响

摘要 目的:研究尿中血红蛋白(Hb)对琼脂糖凝胶尿蛋白电泳蛋白区带分析的影响,探讨血尿蛋白电泳的临床价值。方法:采用十二烷基磺酸钠-琼脂糖凝胶电泳(SDS-AGE)技术对不同程度血尿进行蛋白电泳,以观察Hb带以及尿液中的其他大、中、小蛋白区带的分布。结果:在小分子蛋白区前沿,试带法Hb(+)17例未出现Hb带,Hb(++)、Hb(+++)30例均不同程度地出现Hb带。结论:Hb区带出现在小分子量蛋白区,如将其作为小分子量蛋白百分比报告结果,实为对尿中蛋白质的来源定位错误,可能导致临床误诊。 ObjectiVe To study the effect of urinary hemoglobin (Hb) on agarose gel electrophoresis for urinary protein analysis and its clinical values. Methods SDS-PAGE was applied to detect hematuria at different levels to observe the distribution of Hb band and other large, moderate, and small bands of urinary proteins. Results In the front of small molecular weight protein components, Hb band was not found in seventeen samples of Hb (+) but it was present to different extents in all the 30 specimens of Hb (+ +) or Hb (+ + +). Conclusions Hb band is present in the area of small molecular weight protein components. If this phenomenon is reported as the laboratory result for the percentage of small molecular weight proteins, it really will be a mistake on determining the sources of urinary proteins, which may mislead the clinical diagnosis.
