摘要
目的:探讨更简单快捷的骨髓铁染色方法。方法:试验4%盐酸、40g/L亚铁氰化钾及2者等体积混合液3种方法固定外周血涂片及骨髓涂片的效果。对50例住院患者骨髓涂片分别采用常规方法和省略甲醇固定步骤的简化方法做铁染色,随机双盲观察细胞外铁和细胞内铁。结果:血涂片及骨髓涂片经4%盐酸和40g/L亚铁氰化钾的等体积混合液处理全部固定良好,而经4%盐酸、40g/L亚铁氰化钾作用全部未固定。经秩和检验比较50份骨髓涂片2种方法铁染色外铁及内铁结果无差异。结论:4%盐酸和40g/L亚铁氰化钾的等体积混合液对骨髓涂片除染色作用还兼有优良的固定作用;简化方法突显简单省时、省试剂、少污染的优点,适应临床即时性需求。
Objective:To study a simple method of bone marrow iron staining. Method:Peripheral blood slides and bone marrow slides were fixed by 4% cidum hydrochloricum liquid, or 40 g/L ferrocyanatum Kalium liquid,or mixed liquor with the same volum of these two above, respectively. Bone marrow samples of 50 patients in hostipal were stained by routine method or simplified one which omited the step of mehanol fixation at the same time. Intra-cellular iron and exocellular iron were observed through light microscope. Result:Peripheral blood slides and bone marrow slides all could be fixed by the mixed liquor of 4 % cidum hydrochloricum and 40 g/L ferrocyanatum kalium, but not by 4% cidum hydrochloricum liquid or 40 g/L ferrocyanatum Kalium liquid. Iron deposit stained by routine method and simple one were not different according to rank-sum test. Conclusion: Bone marrow slides can be fixed by the mixed liquor of 4% cidum hydrochloricum and 40 g/L ferrocyanatum Kalium. Simplified staining method for iron deposit in bone marrow makes some prominent advantages including being more simple, saving time and agent with less pollution. It suits for the instantaneous need in clinic.
