摘要 [目的]研究影响西洋杜鹃离体试管苗开花的因素。[方法]以西洋杜鹃无菌试管苗为材料,研究培养基、外源激素、外植体生理状态、光照、活性炭对西洋杜鹃离体试管苗开花的影响。[结果]低盐量、低硝酸盐的1/2 Read培养基较适合西洋杜鹃花芽的诱导,平均诱导率为34.44%;保持一定湿度的含0.7%琼脂的半固体培养基诱导花芽的效果好。IBA 2.0 mg/L是诱导花芽产生的最适宜激素用量。茎尖分化的试管苗最适宜作花芽诱导的材料。光强强度3 000 lx,光照16 h/黑暗8 h的光照条件适合花芽的诱导培养。培养基中加入0.2%活性炭能显著提高花芽诱导率。[结论]外植体生理状态、培养基成分及状态、光照强度和时间、植物激素等对西洋杜鹃离体培养试管苗开花均有影响。 [ Objective]The objective of the study was to reveal the several factors influencing flowering of test-tube plantlets cultured in vitro of Rhododendron hybridum. [Method] With the shoots culture in vitro as explants, the factors influencing the flowering such as medium, phytohormone, physiological state of explants, illumination, activated carbon were hwestigated. [ Result] The 1/2 Read medium with 2.0 mg/L IBA, 0.7% agar, 0.2%activated carbon was suitable for inducing flower buds and the mean induction rate was 34.44%. The shoots derived from stem-tip was best material to induce flower bud. Illumination intensity of 3 000 Ix, photoperiod of 16 h, temperature of 28 ℃ were suitable for the induction of flower bud. [ Conclusion] The physiological state of explants, medium components and state, phytohormone, illumination intensity and photoperied, activated carbon etc. all bad an influence on flowering of test-tube plantlets cultured in vitro of Rhododendron hybriclum.
