S179C突变提高字佐美曲霉木聚糖酶(XynⅡ)热稳定性的研究 被引量:4

Enhanced Thermal Stabilization of Aspergillus usamii Family-Ⅱ Xylanase by S179C-directed Mutagenesis

摘要 宇佐美曲霉木聚糖酶(XynⅡ)是G/11族高比活木聚糖酶的一种,通过S179C定点突变,发现酶最适反应温度由原来的50℃提高到52℃;50℃热处理,半衰期由原酶的23 min延长到突变后酶的70 min,热稳定性提高到原来的3倍;最适反应pH无变化。 Xylanase from Aspergillususamii belongs to family-11 and is a type of xylanase that has high relative activity. Utilizing the method of S179C-directed mutagenesis, we improved its optimum temperature from 50℃ to 52℃. It maintained 50% residual activity after 70 min of incubation at 50℃, while the residual activity of the native xylanase was 50% only after 23 min at the same incubation temperature. The themostability was enhanced by 3-fold. Optimum pH value, however, did not present notable changes
