摘要
[目的]利用RAPD技术对草鱼、鲢鱼和鲫鱼群体遗传变异进行初步研究。[方法]用20种10 bp长的随机引物对草鱼、鲢鱼和鲫鱼进行RAPD-PCR扩增,从20种引物扩增出来的DNA条带中筛选条带清晰的6种引物,分析这3种鱼的遗传变异。[结果]结果表明:草鱼与鲢鱼的RAPD指纹图谱带型差别不明显,草鱼与鲢鱼、鲫鱼品种间的带纹相似系数分别为0.375和0.416,遗传距离分别为0.625和0.584;鲢鱼与鲫鱼品种间带纹相似系数为0.366,遗传距离为0.634。[结论]RAPD是一种非常灵敏的种间鉴定技术,特别是对鱼种类的鉴定有重要意义。
[ Objective ] The research aimed to study the population genetic variation of Ctenopharyngodon iddlus, Carassius auratus, Hypoph- thalmichthy molitrix with RAPD, [ Method] Twenty 10bp primers were applied and six primers were found from the primers by screening the amplified clear bands. [ Result ] The results indicated that RAPD patterns of grass carp had no remarkable difference between those of crucian carp. The band sharing between grass carp and the two carps (crucian carp and silver carp)were 0. 375 and 0. 416,and the corresponding genetic distances between them were 0. 625and 0. 584. The band sharing between silver carp and crucian carp was 0. 366 and genetic distance was0. 634. [ Conclusion ] RAPD was a powerful technique for species indentification, especially for fish.
