摘要 研究HbA1C形成过程,结果表明该过程遵守化学热动力学一般规律:pH影响第一步反应,最适pH为8.0;温度、反应时间影响第二步反应,最适温度为40℃,饱和反应时间是100 h左右;体外诱导HbA1C形成,葡萄糖浓度可达5%,同时对某些有机物对HbA1C形成的影响也进行了讨论.研究发现HbA1C氧解离曲线左移及"S"形弧度变小.制定了诱导提高及纯化HbA1C的方案,纯化的HbA1C相对Hb回收率是22%,跟体内自然形成的HbA1C有相同的免疫原性. The study on HbA1C formation process showed the reaction complied with kinetics law: pH influenced the first step of reaction, and the most suitable value is 8.0; the temperature and the reaction time influenced the second step, and the most suitable temperature is 40 ℃, the saturated reaction time is 100 hours. The concentration of glucose used for inducing HbA1C in vitro can even reach 5%. In addition, some medical organic compounds which effect HbA1C formation have being studied. The research of HbA1C showed that the blood oxygen dissociation curve moved to the left side and curve "S" radian waned. The schedule for HbA1C purification has been made, and the recovery of purified HbA1C to Hb was 22 %, which had the same immunogenicity with that formulated naturally in vitro.
