汉黄芩素对移植人卵巢癌SKOV3细胞裸鼠组织端粒酶蛋白hTERT影响与凋亡相关性研究 被引量:4

Study on the Effect of Wogonin on Telomerase Gene of Nude Mice with Transplanted Human Ovarian Cancer SKOV3 Cells and its Relationship with Apoptosis

摘要 目的评价汉黄芩素对移植人卵巢癌SKOV3细胞裸鼠组织端粒酶蛋白hTERT影响与凋亡相关性。方法移植人卵巢癌SKOV3细胞的裸鼠随机分为5组:汉黄芩素高剂量组,低剂量组,空白对照组,常规化疗组(顺铂)和联合用药(顺铂+汉黄芩素)组。采用抗生素-生物素过氧化酶复合物(S-P)免疫组化法检测裸鼠组织中hTERT,Bc l-2,Bax蛋白及凋亡指数的表达。结果裸鼠组织经汉黄芩素作用后,随着汉黄芩素剂量的增大,对瘤体端粒酶hTERT蛋白表达有抑制作用,呈剂量依赖性。Bc l-2蛋白表达逐渐下调,与凋亡指数变化的表达呈相反变化趋势,而Bax蛋白表达逐渐上调,与凋亡指数变化的表达呈同向变化趋势。结论汉黄芩素在体内具有抗肿瘤和抑制端粒酶活性的作用,抑制强度与剂量成正比,下调端粒酶活性,同时也伴随细胞的凋亡,可能是药物对端粒活性的抑制和对端粒结构的破坏,启动了凋亡基因。 Objective Regulating telomerase activation is an ideal way to treat cancer . Current studies show that wogonin has the attributes of antioxidation and inhibition on the growth of tumor cells . This study aimed at evaluating the relationship between telomerase activation and apoptosis after wogonin administration in nude mice which implanted human ovarian cancer cells SKOV3. Methods Nude mice with implanted human ovarian cancer cells SKOV3 were randomly divided into five groups, including high dose group ,low dose group , normal control group , cisplatin therapy group, and combined therapy group (cisplafin and wogonin) . The expression of hTERT, Bcl -2, Bax and the apoptosis index were detected by immunohistoehemistry. Results The results showed that wogonin could significantly suppress the growth of tumor compared with controlled group. The down expression of telomerase activation and the effectiveness was related to the dose of wogonin . With the increase of suppressing on tumor , the expression of Bcl - 2 decrease regularly , and it showed the opposite tendency of the apoptosis index . While the expression of Bax increased regularly , and it showed the same tendency with the change of apoptosis expression. Conclusion Wogonis can suppress the growth of tumor and regulate telomerase activation. The effectiveness is related to the dose of wogonin , the effect of wogonin would be better if it is combined with eisplatin in the treatment of tumor . It downregulates the activation of telomerase accompanied with apoptosis of cells , the possible reason is that it damages the structure of telomerase and switch on apoptosis gene .
