青钱柳不同外植体组织培养及褐变防止的研究 被引量:21

摘要 目的探索青钱柳快速繁殖途径和褐变的防止方法。方法分别在不同季节采取青钱柳的嫩芽、带节茎段和嫩叶作为外植体,对外植体进行4种不同方式的处理后,接种到不同激素种类和激素组合的培养基中进行培养。结果春季取材更易诱导出愈伤组织和新芽;诱导茎段出芽的最佳培养基为B5+BA3.0 mg/L+NAA0.1 mg/L+IBA0.01 mg/L;适宜于嫩叶、嫩芽愈伤组织诱导的培养基分别为:B5+BA5.0 mg/L+NAA0.1 mg/L+IBA0.1 mg/L和B5+2,4-D1.0mg/L+BA0.1mg/L;外植体先于4℃低温预处理5 d和直接用烧红的手术刀快速切割外植体后再进行接种可有效减少外植体的褐变。结论青钱柳组织培养可诱导芽的产生和愈伤组织的形成。 Objective To study the rapid propagation and prevention of browning of Cyclocarya paliurus( Batal. ) Ijinskaja. Methods Tender buds, nodular stem segments, and tender leaves were taken as explants in different seasons . The explants were treated in four different ways, and then cultured on different mediums. Results Spring was the best season for collecting and the most suitable medium for nodular stem segments to induce buds was B5 + BA3.0 mg/L + NAA0.1 mg/L + IBA0.1 mg/L;as for tender leaves , B5 + BA5. 0 mg/L + NAA0.1 mg/L + IBA0. 1 mg/L was optimum to induce callus, and B5 + BA1.0 mg/L + NAA0.01 mg/L + IBA0.1 mg/L was the best for tender buds to form callus. Putting the explants under the condition of 4℃ for 5 days and cutting the explants quickly with a aglow scalpel before inoculating could decrease the browning effectively. Conclusion The tissue culture of Cyclocarya paliurus ( Batal. ) Ijinskajacan can induce buds and callus .
