皂角刺皂苷对大鼠前列腺增生模型影响的实验研究 被引量:7

摘要 目的探讨中药皂角刺提取物皂角刺皂苷对前列腺增生(BPH)的影响及作用机制。方法将40只SD大鼠去势7d后皮下注射丙酸睾酮5 mg/kg,同时实验组灌胃给予皂角刺皂苷溶液4,2 mg/kg,癃闭舒组灌胃给予癃闭舒胶囊100 mg/kg。于给药30 d后处死,观察前列腺重量和组织结构改变,并通过免疫组化法检测PCNA和bFGF表达的变化。结果实验组大鼠前列腺重量、PCNA指数及bFGF表达水平均低于模型对照组,其中尤以皂角刺皂苷大剂量组最显著(P<0.01)。皂角刺皂苷大剂量组与癃闭舒组比较差异亦有显著性,而皂角刺皂苷小剂量组与癃闭舒组比较无显著差异。结论皂角刺能明显抑制模型大鼠的BPH,其机制可能是通过抑制前列腺细胞的增殖及减少前列腺组织中bFGF的表达而实现的。 Objective To study the effect of Chinese Honeylocust Spine on benign prostatic hyperplasia (BPH) and its mechanism. Methods Forty ovariectomized SD rats were administered testosterone propionate 5mg/kg subcutaneously, At the same time, saponin high and low dose groups were treated respectively with saponin fluid 4mg/kg and 2mg/kg by oral administration and Longbishu Capsule(LBS) group with LBS 100mg/kg, The rats were decapitated after 30 days, the weight of prostate and lightmicroscopic examination were undertaken, PCNA index and expression of bFGF were analyzed by means of immunochemical technology, Results The weight of prostate, PCNA index and the content of bFGF in three medicated groups were significantly less than those of the model group, especially in saponin high dose group. The difference between saponin high dose group and LBS group was also remarkable, But it was not between saponin low dose and LBS group, Conclusion Saponin could suppress the benign prostatic hyperplasia, The mechanism might be the inhibition of cell hyperplasia of prostate and the expression of bFGF.
