摘要
目的探讨Fractalkine在下肢静脉性溃疡发病机制中的作用。方法采用逆转录-聚合酶链反应(RT-PCR)和免疫组织化学技术对Fractalkine在慢性静脉功能不全患者、静脉性溃疡患者和正常人下肢静脉血中的mRNA表达水平进行半定量检测,对其在手术前后溃疡及其周围局部皮肤中的表达进行定性和定位检测。结果Fractalkine mRNA表达在静脉性溃疡患者较单纯静脉功能不全患者和正常人明显增高(P〈0.05);手术前溃疡边缘组织的Fractalkine的表达显著高于正常皮肤组织(P〈0.05),手术后愈合中或愈合后的溃疡组织Fractalkine的表达显著低于手术前溃疡边缘组织(P〈0.05)。结论Fractalkine的表达与下肢静脉性溃疡的发病有密切关系,Fractalkine的高表达可能是导致肢静脉性溃疡的发病的重要因素。
Objective To study the role of Fractalkine in the pathogenesis of venous ulceration. Methods The patients were divided into ulceration group, non- ulceration group and control group. The expression of Fractalkine mRNA in bloed was detected by reverse transcription-polymerase chain reaction (RT-PCR). The expression of Fractalkine was detected in normal skin ,and the tissue at the margin of ulceration and postoperative cured or curing ulceration by immunohistochemical staining. Results In ulceration group, Fractalkine mRNA expression in blood was increased obviously as compared with the other two groups (P 〈 0.01 ). There was significant difference in the expression of Fractalkine between normal skin and the tissue at the margin of ulceration, and also between the tissue of cured or curing ulceration and the tissue at the margin of ulceration. Conclusion The high expression of Fractalkine is closely related with venous ulceration and may be one of the important mechanisms of venous ulceration.
