摘要
采用不同比例和不同N含量的MS培养基,对蕨菜孢子萌发后的原叶体进行培养,以观测最适宜原叶体生长和受精的MS培养基比例及N浓度。试验结果表明,1/2MS培养基为促进原叶体受精的最适培养基;全MS培养基对前期孢子萌发效果最好,但后期不利于受精。N含量为4/5的MS培养基最有利于原叶体受精;全N含量的MS培养基致使原叶体营养生长过旺,不利于受精;而N含量较低的MS培养基致使原叶体生长瘦弱,受精效果也较差。
The spores of bracken [Pteridium aguilinum (L.) Ruhn var.] were cultured in MS medium which varied in MS medium ratio and nitrogen content to examine the optimum ratio of MS medium and the nitrogen concentration for spore germination and prothallus fertilization. The result indicated that 3/5 MS medium was the optimum ratio for fertilization; the MS medium made spore germinate well during the early stage but went against fertilization in later stage. The optimum concentration for prothallus fertilization was 4/5 nitrogen of the whole MS medium. However, higher nitrogen concentration made an excessively prosperous vegetative growth which was harmful for fertilization, lower nitrogen concentration made an emaciated vegetative growth which was also harmful for fertilization.
