摘要
目的:研究高良姜素诱导肝癌细胞系BEL-7402细胞凋亡。方法:MTT法测定BEL-7402细胞毒性及细胞生长,荧光显微镜观察细胞形态学的变化,流式细胞仪分析细胞周期和线粒体膜电位的变化。发色底物法检测Caspase-3、Caspase-6和Caspase-9活性变化。结果:高良姜素对BEL-7402细胞IC50为30.15mg/L,BEL.7402细胞生长曲线表明,高良姜素浓度增高,生长率明显下降。细胞凋亡可在20~80mg/L高良姜素处理后24h出现。凋亡细胞主要表现为核染色质固缩,荧光染色增强。高良姜素阻断细胞于G1期,线粒体膜电位降低。Caspase-3、Caspase-6和Caspase-9被激活,呈时间依赖性改变。高良姜素处理BEL-7402细胞6h时Caspase-9活性最高,而Caspase-6活性在12h达峰值,Caspase-3活性峰值时间在18h。结论:高良姜素可以诱导BEL-7402细胞发生凋亡,其途径可能是通过线粒体旁路。
Objective: To investigate apoptosis of BEL-7402 cells induced by Galangin. Methods: Cytotoxicity and cell viability were assayed by M3T methods. Morphologic assessment of apoptosis was performed by fluorescence microscope, cell Cycle and mitochondrial membrane potential were analyzed by flow cytometry, Caspases Acitivities were detected by chromogenic substrate assay. Resuits: BEL-7402 ceils treated with Galangin showed apparently cytotoxicity, ICs0 of Galangin was 30. 15 mg/L. The growth curve showed that the ratio of growth decreased with the increasing of concentration of Galangin. The apoptosis appeared 24 hours after treated with 20- 80 mg/L Galangin, morphologic changes included nuclear chromatin condensation and fluorescence strength was observed with fluorescence microscope. The activity of Caspase-9 attained to peak value at 6h, while Caspase-6 activity was up to peak value at 12h and Caspase-3 activity attained to peak value at 18h. Conclusion: Galangin may induce BEL-7402 ceils apoptosis by mitochondrial pathway.
