摘要
根据化学动力学原理,采用单一浓度,间歇式反应器,利用酶促反应对反应器中底物浓度的自动耗减特性进行了酶动力学研究,并以小麦的RuBP羧化酶为例测定出Km(CO_2)=14.4μmo1.1^(-1),Vmax=516μmol CO_2.mg_(-1)Ch1.h^(-1),与前人用常规方法测定的结果基本一致,但与常规方法相比,此方法可以大大简化测定手续,减少系统误差。
Enzymatic kinetics was studied chemokinetically by using an in-termittent reactor with single substrate concentration , and depletion ofenzymatic reaction to substrate concentration in the reactor. RuBPC ofwheat (Triticum aestivum) was studied with the new way. It was found thatK_m(CO_2)=14.4μmol·1^(-1), V_(max)=516·mol CO_2·mg^(-1)·chl·h^(-1). The results areconsistent with published data. The new method is simpler and makes less errorthan the old ones.
