摘要
目的观察骨髓间充质干细胞(BMSC)移植后对阿霉素心肌病心脏功能的影响。方法健康日本大耳白兔随机分为3组:①细胞移植组(n=10);②生理盐水注射组(n=10);③正常组(n=7)。前2组动物用盐酸阿霉素静脉注射(1 mg/kg 2次/周共8周)诱导阿霉素心肌病模型。分离培养BMSC,模型诱导成功后的第2周采用直接注射途径将BMSC移植到心肌内,移植后2周评价各组动物的心脏结构和功能,并用HE染色观察阿霉素心肌病模型的心肌细胞的病理改变。结果静脉注射阿霉素可以引起心肌细胞的肥大,空泡变性以及坏死,导致心腔扩大、心脏功能下降。细胞移植2周后,细胞移植组与生理盐水组相比心脏功能有明显的改善,EF、LVAW、LVPW、LVSD、LVDD明显提高(P≤0.05);LVDP、HR显著降低(P≤0.05)。结论BMSC移植到阿霉素性心肌病模型中能改善受损的心脏功能。
Objective To investigate the effects of BMSC implantation on cardiac function of rabbits cardiomyopathy model. Methods Twenty seven rabbits were randomized into 3 groups: cardiomyopathy group, cell implantation group and sodium chloride implantation group. Adriamycin was applied intravenously for 8 weeks to create rabbits cardiomyopathy model. Two weeks after the final adriamycin injection, rabbits were transplanted intramyocardialy with BMSC or saline. Two weeks after transplantation, echocardiography and histology analyses were conducted. Results Two weeks afer cell transplantation, compared with the saline group, heart function was significantly improved. Pathological examination demonstrated that there were viable transplanted cells in the transplanted hearts. Conclusion BMSC can improve heart function in rabbits cardiomyopathy model.
