离心运动大鼠骨骼肌微损伤组织形态学变化及人参G115的干预 被引量:4

摘要 背景:动物和人体实验结果证明人参及其活性成分能提高机体的运动能力。目的:通过运动性骨骼肌微损伤动物模型,从形态学角度拟验证人参G115对离心运动后骨骼肌微损伤的修复及营养干预作用。设计、时间及地点:随机对照动物实验.于2005-01在国家体育总局运动医学研究所运动营养研究实验室完成。材料:成年SPF/VAF级雄性SD大鼠72只,体质量270~300g:人参G115为浙江霍夫曼德公司产品。方法:SD大鼠按体质量层次随机分组3组,安静组、对照组和实验组。以大鼠离心运动为模型,实验组大鼠以16m/min的速度,坡度为-16°运动5min,休息2min,总运动时间为120min。实验组和对照组动物每天分别灌胃1.5%人参G115溶液2札和等量蒸馏水,于运动后即刻,24,48和72h,每组取8只观察各项指标。主要观察指标:各组骨骼肌组织细胞微细结构和超微结构及结蛋白和波形蛋白的表达。结果:离心运动后骨骼肌组织形态结构和超微结构都发生了不同程度的变化.以运动后24~48h间最为严重;实验组运动后72h微损伤能明显修复。离心运动后出现抗结蛋白抗体染色脱染和抗波形蛋白抗体染色阳性现象,以运动后24h最严重:运动后即刻、24h、48h,实验组骨骼肌结蛋白脱染细胞百分数比对照组下降,各时间点波形蛋白的免疫反应分值上升。结论:人参Gl15对运动引起的微损伤有明显的抑制作用,使运动后微损伤出现的程度降低,对离心运动引起的结蛋白表达下降和波形蛋白的表达也有明显的抑制作用。 BACKGROUND: Ginseng and its active components can enhance the exercise ability of organism through animal and human trials. OBJECTIVE: To verify the effect on the repair of skeletal muscular micro-injury and the nutrition intervention through morphometric methods in animal models supplied with ginseng G 115 after eccentric exercise. DESIGN, TIME AND SETTING: Randomized control animal experiments were carried out in the Laboratory, Sports Nutrition Center, National Research Institute of Sports Medicine, General Administration of Sports of China in January 2005. MATERIALS: Seventy-two male SD rats of SPF/VAF grade, weighing 270-300 g, were adopted; ginseng G115 was produced by Zhejiang Hoffmann Company (China). METHODS: According to the weight, 72 rats were randomly divided into rest group, control group and exercise groups. Rat models of eccentric exercise were induced. Rats of the exercise group were subjected to the exercise according to -16° gradient at a speed of 16 m per minute for 5 minutes, with the interval of 2 minutes, totally 120 minutes. Exercise group and control group were administrated intragastrically with 1.5% ginseng G115 solution (2 mL) and equal volume of distilled water, respectively. Post-exercise 0, 24, 48 and 72 hours, eight rats from every group were observed. MAIN OUTCOME MEASURES: Microstructure and ultrastructure of skeletal muscles were observed, as well as the expression of desmin and vimentin. RESULTS: Subsequent to eccentric exercise, the microstructure and ultrastructure of skeletal muscles were changed by different degrees, especially during post-exercise 24-48 hours; The micro-injury of the exercise group could be repaired at post-exercise 72 hours. Anti-desmin antibody and anti-vimentin antibody were detected to be positive for the stain, which was the most serious at post-exercise 24 hours. Compared to the control group, the percentage of the cells negative for desmin stain descended, while immune reaction value of anti-vimentin antibody hoisted in every time point post-exercise. CONCLUSION: Ginseng G115 can obviously restrain the micro-injuries caused by the exercise and reduce the injury degree after exercise. Ginseng G115 also decreases the expression of desmin and vimentin.
