摘要
目的:观察尼莫地平(Nimodipine)和山莨菪碱(654-2)预处理在大鼠肝缺血再灌注损伤过程中对心脏保护作用机制的不同点。方法:32只健康Wistar大鼠分为对照组、缺血再灌注4 h组(I/R4 h组)、654-2干预组(I/R4 h+654-2)和尼莫地平干预组(I/R4 h+N)。观察测定心率(HR)、左室收缩压(LVSP)、左室舒张末期压力(LVEDP)以及心肌组织丙二醛(MDA)、髓过氧化物酶(MPO)的变化。结果:I/R4 h组LVSP较对照组明显下降(P<0.05),I/R4 h+654-2组和I/R4 h+N组较对照组和I/R4 h组明显升高(P<0.01);LVEDP在I/R4 h+654-2组和I/R4 h+N组明显低于对照组和I/R4 h组(P<0.01);I/R4 h组MDA的含量明显增加,与对照组比较差异有统计学意义(P<0.01),I/R4 h+654-2组I、/R4 h+N组与对照组和I/R4 h组比较差异有统计学意义(P<0.01);I/R4 h+654-2组与对照组比较MPO明显升高(P<0.01),与I/R4 h组比较差异无统计学意义(P>0.05),I/R4 h+N组与对照组和I/R4 h组比较,MPO的差异有统计学意义(P<0.01)。结论:肝缺血再灌注损伤致心功能改变的过程中,尼莫地平预处理对心功能的保护作用强于山莨菪碱。
Objective： By using Nimodipine and 654-2 pre-treat during the process of liver ischemia-reperfusion injury, to investigate the mechanism of the heart-protection function. Methods： Thirty two healthy wistar rats were randomly divided into 4 groups： control group （CTL）, 4 hours reperfusion following ischemia for 30 min （I/R4 h）, 654-2 pre-treat group （I/R4 h＋654-2）, Nimodipine pre-treat group （I/R4 h ＋N）. To observe the change of those indexes： HR, LVSP, LVEDP, MDA, MPO. Results： LVSP was obviously decreased in I/R4 h group （P 〈0.05）, but it obviously increased in I/R4 h＋654-2, I/R4 h＋N comparing to CTL and I/R4 h group （P 〈0.01） ; LVEDP obviously decreased in I/R4 h＋654-2 and I/R4 h＋N compared to CTL and I/R4 h group （P 〈0.01） ; the level of MDA obviously increased in I/R4 h group, there was significant difference compared with CTL （P 〈0.01）, there was also an obvious significant difference in I/R4 h＋654-2 and I/R4 h＋N compared to CTL and I/R4 h group （P 〈0.01） ; the level of MPO obviously increased in I/R4 h＋654-2 （P〈0.01）, but there was no significant difference compared with I/R4 h group （P 〉0. 05）, there were significant differences between I/R4 h＋ N and CTL, I/R4 h group （P 〈0.01）. Conclusion：During the process of dysfunction of heart induced by liver ischemiareperfusion injury, the protective effect of Nimodipine to the heart is better than that of 654-2.
