摘要
以幼小仓鼠肾细胞(BHK21)为研究对象,将DMEM培养基中血清体积分数从15%降至1%进行驯化培养。比较驯化过程中细胞的生长变化;同时测定血清体积分数由15%降至2%的过程中,培养液中葡萄糖、谷氨酰胺等氨基酸以及乳酸、氨的浓度变化。结果表明,血清体积分数由15%降至2%,细胞生长的延迟期延长96 h;同时细胞对葡萄糖、谷氨酰胺的代谢速率以及乳酸的积累速率都降低了50%,而氨的积累速率降低了29%。
Adaptation of baby hamster kidney(BHK21)cells to culture medium DMEM with a reduced new calf serum (NCS) level from 15% to 2% was investigated. Growth rate and primary metabolism of 13HK21 cells adapted to grow in different level serum-supplemented DMEM were compared as well. The results showed that with decreasing serum volume fraction, the lag phase of cell growth cycle became longer. Regarding to primary metabolism of BHK21 cultured in lower serum level medium, consumption rates of glucose, glutamine and accumulation rate of lactic acid decreased 50 % ,meanwhile, accumulation rate of ammonia decreased 29%.
