摘要
目的:了解烧冲复合伤早期肺组织细胞间粘附分子-1(ICAM-1)的表达变化规律及其与多形核白细胞(PMN)在肺内聚集的关系.方法:测定肺组织髓过氧化物酶(MPO)以反映PMN肺内聚集的情况;用地高辛标记ICAM-1cDNA探针进行肺组织原位杂交;用SP法进行肺组织ICAM-1免疫组化染色.结果:各致伤组动物伤后均有不同程度的肺组织MPO活性增高,其中以复合伤组最明显,肺组织ICAM-1mRNA及其蛋白质表达均较对照组显著增多,以伤后6小时最为明显.结论:烧冲复合伤早期肺ICAM-1增多可能与PMN的肺内聚集有关.
Aim: To realize changes of pulmonary intercellular adhesion molecule 1(ICAM 1) expression and accumulation of pulmonary polymorphonuclear neutrophil (PMN) in the early stage of burn blast combined injury. Methods : Pulmonary myeloperoxidase activity was assayed to show accumulation of PMN in the lung. Lung tissue in situ hybridization was determined by digoxin labeled ICAM 1 cDNA probe. Immunohistochemistry of ICAM 1 in the lung was measured. Results: Pulmonary myeloperoxidase activity of each injured group increased after injury. The most obvious changes was showed in burn blast combined injury. The expression of ICAM 1 mRNA and its protein of the lung in each injured group were markedly increased than that of the control. The peak level was observed at 6 hours after injury. Conclusion: The process of accumulation of PMN in the lung might be related to the increase of ICAM 1 expressing in the lung at the early stage of burn blast combined injury.
