摘要
目的研究罗格列酮对糖尿病大鼠肾组织氧化应激的影响。方法36只雄性Wistar大鼠随机分为正常对照组(NC)、糖尿病组(DM)、糖尿病罗格列酮治疗组(DR)。腹腔注射链脲佐菌素(STZ)诱导糖尿病模型,罗格列酮[5mg/(kg·d)]对DR组进行治疗性灌胃。8周末称取体重、肾重、肾重/体重,检测尿白蛋白排泄率(UAER)及尿肌酐;测定各组生化指标包括血糖(BG)、胆固醇(TC)、三酰甘油(TG)、血清肌酐(SCr);检测肾组织与尿中丙二醛(MDA)的含量及肾组织超氧化物歧化酶(SOD)、过氧化氢酶(CAT)与谷胱甘肽过氧化物酶(GSH-Px)活性;PAS染色作肾组织切片分析肾小球面积、肾小球体积及系膜区面积。结果8周时,与DM组比较,DR组大鼠BG、TC、TG差异无显著性。DM组大鼠肾重、肾重/体重、UAER、Ccr、肾小球面积、肾小球体积、系膜区面积较NC组均明显增加,DR组上述改变较DM组均明显减轻(P<0.05)。DR组肾组织及尿中MDA含量明显低于DM组,SOD、CAT、GSH-Px活性明显高于DM组(P<0.05)。结论罗格列酮对糖尿病大鼠肾组织有明显保护作用,其机制部分可能与抑制肾组织氧化应激有关。
[Objective] To study the effect of Rosiglitazone on the oxidative stress in the kidney in diabetic rats. [Methods] 36 Wistar rats were randomly divided into three groups: NC group, normal control group. DM group, diabetes meUitus group. DR group, Rosiglitazone-treated diabetes mellitus group. Diabetic rats were induced by streptozotocin (STZ), and the rats in DR group received Rosiglitazone [5 mg/(kg·d)] through stomach perfusion. The following parameters were measured after 8 weeks:body weight and kidney weight, urine albumin excretion rate (UAER) and urine creatinine,biochemical indexes including blood glucose(BG), total cholesterol(TC), triglycerides (TG), serum creatinine(SCr), the activities of antioxidant enzymes including superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH - Px) and maleic dialdehyde (MDA), renal morphology parameter including glomerular area, glomerular volume and mesangial area by PAS staining. [Results] After 8 weeks, There were no significant difference in BG, TC, TG levels between DM and DR groups. Increased kidney weight, ratio of kidney weight to body weight, UAER, Ccr and glomerular area, glomerular volume, mesangial area on histological examination of the kidney significantly attenuated by treatment with Rosiglitazone (P 〈0.05). Levels of MDA were higher and SOD, CAT as well as GSH - Px activities were significantly lower in kidney tissue in DM group rats than those in NC group. Rosiglitazone administration could all remit these change (P 〈0.05). [Conclusion] The mechanism of protective effect of Rosiglitazone on kidney may be at least partly correlated with the suppression of increase in oxidative stress in diabetic rats.
