摘要
目的观察白细胞介素12/信号传导子及转录激活子4(IL-12/STAT4)信号途径激活对体外培养的人脐动脉平滑肌细胞(vascular smoothmus clecells,VSMC)增殖的影响。方法不同浓度(2.5、5、10和20ng/mL)IL-12刺激后,用MTT法、Hoechst 33258染色法观察VSMC增殖和凋亡情况,RT-PCR、Western blot分别检测STAT4的mRNA表达和STAT4、磷酸化STAT4(P-STAT4)的蛋白表达变化。结果IL-12呈剂量和时间依赖促VSMC增殖,同时抑制VSMC凋亡。RT-PCR检测发现STAT4的mRNA表达随IL-12浓度增高而增加、Western blot检测发现STAT4、P-STAT4的蛋白表达随IL-12浓度增高而增加。结论IL-12可促进VSMC增殖,该作用可能与激活IL-12/STAT4途径有关。
[Objective] To investigate the effect and molecular mechanism of Interleukin-12 (IL-12) on proliferation of vascular smooth muscle cells(VSMCs). [Methods] VSMCs were treated with IL-12(2.5, 5, 10 and 20 ng/ mL). The proliferation of cells following IL-12 stimulation was estimated by MTT assay, Apoptosis was assessed through Hoechst 33258 staining. The mRNA expression of STAT4 in cells was determined by RT-PCR, the level of both STAT4 protein and phosphorylated STAT4 protein in cells were analyzed by Western blot. [Result] IL-12 significantly promoted proliferation of VSMC in a dose- and time- dependent way while inhibited apoptosis of VSMC, The mRNA expression of STAT4 was increased and correlated with IL-12 concentration. The level of both STAT4 protein and phosphorylated STAT4 protein were increased and correlated with IL-12 concentration. [Conclusion] IL-12 may promote proliferation of VSMC through IL-12/STAT4 signaling pathway.
