摘要
给肝癌大鼠注射济南假单胞菌苗,连续12周.分别于第8、12、16周从同只大鼠体内分离出肝枯否细胞,腹腔巨噬细胞,肺巨噬细胞和血液单核细胞.并分别与人肝癌细胞联合培养,测定5种效应细胞对肝癌细胞的抑增殖作用.结果显示:5种效应细胞对肝癌细胞均具有自然增殖作用,作用强度依次为枯否细胞、腹腔巨噬细胞、脾巨噬细胞、血液单核细胞.在体应用济南假单胞菌苗后,5种效应细胞的抑肝癌细胞作用均显著增强,抑制率分别提高了59.2%~174.2%不等.在体实验为临床肝癌免疫治疗应用济南假单胞菌苗的可能性提供了实验依据.
The rats with hepatoma induced by diethylnitrosamine(DEN) were treated by Pseudomonas Jinanesis Vaccine (PJV). Kupffer cells (KC), peritoneal monocytes(PM) , alveolar macrophages (AM), spleen macrophages (SM) and monocytes of each hepatoma rat were isolated at 8th, 12th and 16th week, and co-cultured with human SMMC-7721 hepatoma cells. Through measuring 3H-TDR incorporation into tumor cell DNA respectively, the cytostatic activities of mononuclear phagocytes on human hepatoma cells were assayed. Five kinds of mononuclear phagocytes possessed different cytosatic activities on hepatoma cells. Among them the effect of KC was the strongest. After treatment with DEN, the cytostatic activities of all mononuclear phagocytes increased obviously by 59. 2%~174. 20% respectively. The results indicated that PJV may be used as the immune stimulant to patients with hepatoma.
