摘要
目的:研究丹参酮B钠盐对大鼠局灶性脑缺血再灌注损伤的保护作用。方法:应用线栓法可逆性的阻塞大鼠大脑中动脉,制备大鼠局灶性脑缺血再灌注损伤模型。复灌24 h后对大鼠进行神经学评分,并测定脑梗死率和含水量。制备脑组织病理切片,观察丹参酮B钠盐对局灶性脑缺血再灌注损伤大鼠脑组织病理学影响;并测定脑组织细胞内Na+,K+-ATPase活力、钙离子含量、超氧化物歧化酶(SOD)活力、丙二醛(MDA)含量,观察丹参酮B钠盐对这些生化指标变化的影响。结果:丹参酮B钠盐能使缺血再灌注后大鼠的神经行为明显改善,显著降低脑梗死率和含水量。脑组织切片病理学检查表明丹参酮B钠盐对脑组织细胞缺血再灌注损伤具有明显保护作用;生化指标测定表明丹参酮B钠盐能降低细胞内钙含量、脑组织MDA含量,增加Na+,K+-ATPase、SOD的活力。结论:丹参酮B钠盐对大鼠局灶性脑缺血再灌注损伤具有很好的保护作用。
Aim: To study the protective effect of sodium tanshinone B( sodium TanB) on focal cerebral ischemia- reperfusion injury in rats. Methods: The rat model of focal cerebral ischemia-reperfusion was established with middle cerebral artery occlusion (MCAO) by reversibly inserting a nylon thread. Neurological scores, infarct size and water content were determined 24 h after MCAO, and at the same time the pathological changes to ischemic cerebral tissues were observed. The contents of intracellular calcium, MDA, the activities of Na^+, K^+ -ATPase and SOD were also measured. Results: Pretreatment with sodium TanB significantly reduced the cerebral infarct size and edema, and produced significant reductions in neurological deficits. Pathological observations proved the protective effect of sodium TanB on neuron cells. The contents of MDA and intracellular calcium were significantly reduced through the pretreatment of sodium TanB. The activities of Na^+, K^+ -ATPase and SOD were also enhanced significantly. Conclusion: Sodium TanB has protective effect against the cerebral ischemia-reperfusion lesion in rats.
