转化生长因子β_1、胰岛素样生长因子1受体在子宫肌瘤中的意义 被引量:4

Expression of transforming growth factor beta-1 and insulin like growth factor-1 receptor in human uterine leiomyoma

摘要 目的探讨转化生长因子β1(TGF-β1)和胰岛素样生长因子1受体(IGF-1R)在子宫肌瘤中的表达及其相关性。方法采用原位杂交方法检测50例子宫肌瘤及瘤旁正常的子宫肌层中的TGFβ1 mRNA的表达,同时采用免疫组化技术检测相应标本中TGFβ1和IGF-1R蛋白的表达。结果(1)TGFβ1 mRNA及蛋白阳性表达均位于细胞浆。在子宫肌瘤中的表达水平分别为1.02±0.38、1.42±0.67,在相应的正常子宫肌壁中的表达水平分别为0.66±0.48、0.58±0.50,两者差异有统计学意义(P<0.01)。(2)IGF-1R在子宫肌瘤和相应的子宫肌壁中的表达水平分别为1.92±0.88、0.80±0.49,两者比较差异有统计学意义(P<0.01)。(3)各指标在增殖期、分泌期肌瘤和肌壁中的表达差异无统计学意义。(4)TGFβ1与IGF-1R无相关性。结论TGFβ1、IGF-1R是子宫肌瘤生长的重要因素。 Objective To investigate the expression of transforming growth factor beta-1 (TGF-β1)and insulin like growth factor-1 receptor (IGF-1R) in human uterine leiomyoma and myometrium. Methods Fifty surgical specimens of uterine leiomyoma and corresponding normal myometrium were collected. The expression of TGF-β1 mRNA was detected by in situ hybridization (ISH); while the expression of TGF-β1 and IGF-1R proteins was detected by immunohistochemical S-P method. Results The positive expression of TGF-β1 mRNA and protein were all located in cytoplasm. The expressive values of TGF-β1 mRNA and protein in uterine leiomyoma were 1.02±0. 38 and 1.42±0.67, respectively,while those in matched normal myometrium were 0.66±0.48 and 0.58±0.50, respectively (P〈0.01). The expressive values of IGF-1R in human uterine leiomyoma and normal myometrium were 1.92±0.88 and 0.80±0.49, respectively (P〈0.01). There were no significant differences in the expression of TGF-β1 mRNA and protein,IGF-1R protein between the proliferative and secretory phases both in uterine leiomyoma and normal myometrium. The expression of TGF-β1 protein was not correlated with that of IGF-1R. Conclusions TGFβ1 and IGF-1R might he important factors for growth of uterine leiomyoma.
