摘要
用组织培养技术获得黄瓜叶片的愈伤组织,进而培养其悬浮细胞,然后用Na+,Ca2+,H2O2分别诱导黄瓜叶片悬浮细胞凋亡,并用显微镜观察和琼脂糖凝胶电泳检测其DNA。Na+(1.4mol/L),Ca2+(500mmol/L),H2O2(10%)作用于黄瓜叶片悬浮细胞后,用显微镜观察到凋亡小体;DNA琼脂糖凝胶电泳检测出凋亡时形成的DNAladder。结果表明:Na+(1.4mol/L),Ca2+(500mmol/L),H2O2(10%)都能诱导黄瓜叶片悬浮细胞凋亡。
The callus of cucumber leaves was cultured in suspension cells and the cells were induced into apoptosis by Na^+, Ca^+ and H2O2. Agarose gel electrophoresis and microscope were carried out to detect DNA fragment and the apoptosis bodies. The results showed that after the treatment of Na^+, Ca^+ and H2O2, at the concentration of 1.4 mol/L, 0.5 mol/L and 10% respectively, the apoptosis bodies and the DNA ladder were detected, implying that the three inducements at the corresponding concentrations could.induce the cucumber callus into apoptosis.
