摘要
目的研究出一种提取骨骼肌肌钙蛋白C(sTnC)的新方法。方法兔骨骼肌经匀浆、离心后上DEAE-Sephadex A25层析柱、Sephadex G75层析柱及Butyl Sepharose层析柱以获得sTnC纯品。结果终产品出现了指状吸收光谱,其吸收峰位于253nm、259nm、265nm、269nm、276nm。采用Diol-150色谱柱进行HPLC分析,出现单一峰。SDS-PAGE分析,得到一条电泳带,计算其分子量为19KD。本实验sTnC的产率为50mg/100g兔骨骼肌,提取周期缩短至6天。结论该方法不仅简化了sTnC的提取纯化步骤,缩短了提取周期,而且使其产率大大提高。
Objective To study out a new method to purify sTnC.Methods Rabbit skeletal muscle was homogenized and centrifuged, then applied to DEAE-Sephadex A25,Sephadex G75 and Butyl Sepharose columns in turn to abtain the purified sTnC.Resuits The final product possessed finger-shape absorbance spectrum which lay in 253 nm.259 nm,265 nm,269 nm、276 nm. Both HPLC and SDS-PAGE showed single band.The molecular weight of the final product was 19 KD and the yield was 50 mg/100 g rabbit skeletal muscle tissue. Conclusion The purification procedure of sTnC was simplified, the cycle was shorter and the yield was higher than previous reported.
