摘要
目的:探讨丝裂霉素对胃癌细胞拓朴异构酶Ⅱα(TOPOⅡα)表达的影响。方法:人胃癌细胞株BGC-823培养,加入丝裂霉素后5h和10h后采集细胞,进行光、电镜观察和免疫组化染色。结果:5h组形态变化不明显,胞核中TOPOⅡα含量减少;10h组,细胞器减少,质膜破裂,其胞质与胞核中TOPOⅡα表达明显减少。结论:丝裂霉素对BGC-823癌细胞有明显的杀伤作用,其途径之一可能与抑制TOPOⅡα基因表达有关。
Objective:To study the killing function of mitomycin(MMC) to gastric cancer cells and the expression of TopoⅡα. Methods:Human gastric cancer cellsBGC-823 was cultured,while MMC was added to culture continuously for 5 h or 10 h. Then the gastric cancer cells were observed under optical microscope and transmission electron microscope and stained with immunohistochemical method as well. Results: The morphology of gastric cancer cell was not changed significantly,but content of Topo Ⅱαin nuclear was reduced in the group for 5 hours. The changes in the group for 10 hours were as follows, the cell organ was obviously destroyed, the cell merabrane were burst,and the expression of TOPⅡα was reduced significantly in cytoplasm and nuclear. Conclusion:Mitomyclin has killing fuction to gastric cancer cells,one of ways was to restrain the expression of TOPO Ⅱα gene.
