摘要
在单因素试验的基础上,利用三因素三水平试验设计结合响应面法对胰蛋白酶水解提取肝素的工艺参数进行优化研究。通过响应面分析建立了回归模型来分析各因素对肝素的提取率的影响,以及个因素间的相互作用。通过典型性分析得到优化肝素提取条件,优化后的工艺参数为:酶水解温度为55℃;酶水解时间2h;料液比1∶15,此条件下肝素得率理论值为211.34mg·kg-1,验证试验肝素得率为206.83mg·kg-1。
Based on the results of single factor experiment, a three-level and three-factor experimental design combined with response surface methodology (RSM) was applied to optimize the trypsin hydrolysis extraction conditions of Heparin. A mathematical model was then developed to show the significant effect of each factor and their interactions on the yield of Heparin. The Reseg Analysis was used to estimated the yield of heparin, the optimized parameters were: enzymic hydrolysis temperature at 55 ~C, enzymic hydrolysis time at 2 hours, and ratio of material solution were 1:15.The theoretic yield of heparin was 211.34mg· kg-1, and under the verificative test condition the practical values was 206.83 mg· kg-1.
