摘要
用超临界CO2流体萃取缫丝蚕蛹油,采用气相色谱-质谱(GC-MS)联用技术分析其脂肪酸组成。测定了3种蛹油样品的碘值、皂化值、酸值、过氧化值等理化指标,研究了缫丝蛹油对四氧嘧啶诱导糖尿病小鼠的降血糖作用。结果表明:从超蛹油中共分离、鉴定出9种脂肪酸,其中以α-亚麻酸、油酸为主的不饱和脂肪酸占脂肪酸总量的72%~75%;以缫丝蚕蛹油灌胃治疗四氧嘧啶诱导的糖尿病小鼠,低、高剂量蛹油组小鼠体重增加,血糖较模型对照组显著降低,降血糖效果与格列苯脲相当;低、高剂量蛹油组糖尿病小鼠胸腺指数和脾指数均比模型组有所提高,小鼠血清及肝脏SOD活性升高,抑制了MDA的产生。超临界CO2萃取的蛹油作为一种富含ω-3型不饱和脂肪酸(α-亚麻酸)的功能性油脂,对开发降血糖新资源功能食品具有一定意义。
Oil samples from residual pupa of silk reeling were extracted with supercritical CO2. The fatty acid compositions of the oil samples were analysed by GC-MS and the iodine value, saponification value, acid value, peroxide value were determined at the same time. Nine fatty acids were separated and identified, there are four saturated fatty acids mainly composed by palmitic acid and stearic acid which account for 27%-28% of the total fatty acids, and five unsaturated fatty acids mainly composed by oleic Acid and α-linolenic acid which account for 72%-75% of the total fatty acids. The hypoglycaemic test result indicated that low dose [300 mg/(kg·d)] and high dose [600 mg/(kg·d)] of pupa oil treatment on diabetic mouse could improve the weights and reduce the blood glucose observably, whose therapy effect was equal to that of glibenclamide. The thymus gland index and the spleen index of the diabetic mouse were improved. The pupa oil may be a good souce of ω-3 unsaturated fatty acid (α-linolenic acid) to explored functional fatty in food and medicine fields.
