摘要 利用鉴定番木瓜雄性性别的特异基因片段(1001bp)的引物和鉴定番木瓜雄性、两性性别的特异基因片段(225bp)的引物,分别以5个品种番木瓜的3种性别植株的总DNA为模板进行多重PCR扩增,结果表明:以雄株总DNA为模板得到1个1001bp和1个225bp的扩增产物,以雌株总DNA为模板没有得到扩增产物,以两性株总DNA为模板得到1个225bp的扩增产物,根据多重PCR扩增结果,可以将番木瓜3种性别植株一次性鉴定出来。 A new rapid way of discriminating male, hermaphrodite and female individuals was developed in papaya based on multiplex PCR reaction. Double pairs of primers were designed: one based on the male specialized sequence with 1 001 bp amplified fragment, and the other based on male and hermaphrodite sequence with 225 bp used in multiplex PCR amplification to genome DNA of male, hermaphrodite and female plains respectively. Two bands (1 001 bp and 225 bp) were amplified in the male plant, and a fragment of 225 bp was amplified only in the hermaphrodite plant, but no bands were amplified in the female plant. This means the three different sex characteristics in papaya could be identified once by the multiplex PCR reaction. This multiplex PCR is quick and accurate and will play an important role in commercial production of papaya.
