摘要 目的观察脂质过氧化反应在酒精导致大鼠急性肝损害中的作用。方法健康雄性Wistar大鼠40只,随机分为4组。A组(空白对照组)灌胃生理氯化钠溶液20 mL.kg-1.d-1;其余各组灌胃等剂量白酒;其中C组(阳性药物干预组1)在B组基础上预先1 h灌胃茴三硫10 mg.kg-1.d-1,D组(阳性药干预组2)在B组基础上预先1 h灌胃硫普罗宁100 mg.kg-1.d-1;实验共10 d。实验第6天、第11天取各组大鼠血液,检测谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)、谷丙转氨酶(ALT)、谷草转氨酶(AST);实验结束时处死动物,取肝组织,HE、Masson染色,光镜检查。结果急性酒精中毒可引起大鼠血清MDA升高,GSH-Px降低;ALT、AST升高,提示有显著的脂质过氧化反应和肝功能损害,肝脏病理出现肝细胞脂肪变性,炎症坏死病变。使用两种药物干预后脂质过氧化反应受到抑制,肝功能和肝脏病理学损害均有减轻,各组间差异有统计学意义。结论脂质过氧化反应在大鼠急性酒精性肝损害的发生中起重要作用,抑制该反应可显著减轻肝脏损伤。 Objective To observe the effect of lipid peroxidation in acute hepatic injury of rat induced by alcohol. Methods Forty male Wistar rats were randomly assigned to four groups. Group A (placebo group) was given orally by saline at the dose of 20 mL·kg-1·d-1, the other groups were 56% alcohol at the dose of 20 mL·kg-1·d-1. Group C(intervention group 1 ) was beforehand administened orally by anethohrithione at the dose of 10 mg ·kg-1·d-1 Group D ( intervention group 2) was beforehand administened orally by tiopronin at the dose of 100 mg ·kg-1·d-1 All rats were treated for 10 days. At the 6th day and the 11 th day of the treatment,the rat bloods were collected to measure alanine amino transaminase ( ALT), aspartate amino transaminase ( AST), glutathione peroxidase ( GSH-Px ), malondial- dehyde(MDA). When the experiment was over,rats were euthanized and the histopathologic alterations were observed. Results At the 6th day and 1 lth day,compared with group A, Group B showed severe hepatic injury with abnormally higher levels of MDA, ALT, AST and the lower GSH-Px, as well as hepatocyte fatty degeneration, even cellular necrosis. The results meant there were obvious lipid peroxidation and injuryed hepatic function. Treatment with above two drugs could inhibit the lipid peroxidation and prevent the rats from acute alcolho-induced hepatic injury. Conclusion The effect of lipid peroxidation is main factor of rat acute hepatic injury induced by alcohol. Inhibiting this effect will light the injury markedly.
