摘要
将苹果杂交种(长富2号×新红星)种子打破休眠,经消毒处理后接种在MS培养基上获得无菌组培苗。以组培苗叶片为试材,通过不同激素组合试验,获得了适宜植株再生的培养基MS+NAA 0.4 mg/L+TDZ 2.0 mg/L。利用含LFY基因的根癌农杆菌EHA105转化苹果叶片,得到转LFY基因的阳性植株,经PCR和RT-PCR检测,有2株出现特异性条带,证明该基因已经整合到苹果基因组中。
Hybrid seeds of apple (Nagafu No. 2 x Starkrirnson) after breaking dormancy were sterilized and vaccinated in MS medium. Leaves of tissue culture seedlings cultured on the medium with different hormone combinations were used to study. The results showed that the optimum regeneration medium was MS + NAA 0.4 mg/L + TDZ 2.0 mg/L. Then using Agrobacterium turnefa- ciens EHA105 containing LFY gene to transform apple leaves, some positive seedlings were obtained, and two of them amplified specific band by detecting of PCR and RT - PCR. It showed that LFY gene had been integrated into the genome of apple.
