附子多糖的提取及免疫活性研究 被引量:26

STUDY ON EXTRACTION OF POLYSACCHARIDE FROM RHIZOMA TYPHONIUM GIGANTEI AND ITS IMMUNOLOGICAL PROPERTIES

摘要 [目的]从白附子片中提取出多糖,并对其免疫活性进行初步研究。[方法]采用水提醇沉法提取附子多糖,Sevag法除蛋白,苯酚-硫酸分光光度法测定多糖的含量。分别用绵羊红细胞(SRBC)和卵清蛋白为抗原给小鼠注射后,将附子多糖分为3个剂量[50100200mg/(kg·d)]灌服小鼠7d,加强免疫后,检测相应抗体生成水平,评价其免疫活性。[结果]附子多糖的含量为(5.431±0.334)%,回收率97.26%,RSD为3.78%,线性范围为5~40μg/ml。中剂量实验组SRBC抗体生成水平(P﹤0.05)和卵清抗体生成水平显著高于对照组(P﹤0.01)。[结论]附子多糖对正常小鼠机体免疫有增强作用。 [Objective] To extract polysaccharide from Rhizoma Typhonium Gigantei, mid to primarily study on its immunity activity. [ Methods ] Polysacchayide was extracted from Rhizoma Typhonium Gigantei by water-extraction-alcohol-precipitation method, performed deproteinization by Scrag, and determined by phenol sulfonic acid spectrophotometry. Mices were injected respectively with SRBC and ovalbumin which were employed as antigens, The doses of Rhizoma Typhonium Gigantei polysaccharide were divided into three dosages [50 100 200 mg/(kg.d)], and given orally for seven days. After the secondary immunization, the level of corresponding antibody was determined and its immunological activity was evaluated. [ Flesults] The content of polysaccharide of samples was (5.431±0.334) %, the average recovery was 97.26%, RSD was 3.78%, and standard curve was at the range of 5--40 ug/ml. The levels of antibodies (anti-SRBC and anti ovalbumin) in median dose group of Rhizoma Typhonium Gigantei polysaccharide were significantly higher than those of control groups. [Conclusion] Rhizoma Typhonium Gigantei polysaecharide could enhance the immune function of normal mice.
