摘要
RNA干扰(RNAi)是真核生物中普遍存在的一种自然现象,利用双链RNA诱发宿主细胞同源mRNA降解,从而抑制特异目的基因的表达,不仅具有理论意义,而且具有实用价值。白血病是造血系统常见的恶性肿瘤,虽然通过联合化疗大多数患者可以获得完全缓解,但是最终不可避免的是多药耐药(MDR)导致化疗的失败。MDR已成为目前治愈白血病的最大障碍。因此,逆转MDR成为白血病治疗亟待解决的问题。本文以白血病MDR机制的阐述为线索,综述了RNAi技术逆转白血病MDR的研究进展。
RNA interferance is a common natural phnomenon in the eucaryote. Using double-stranded RNA to induce degradation of homologous mRNA and thereby suppress the expression of the specific target gene has not only theofical value, but also utility value. Leukemia is a common malignant tumor in the hemopoietic system. Although most patients may acquire complete remission by using combined chemotherapy, multidrug resistance is inevitable finally due to chemotherapy failure. At present, multidrug resistance has become the biggest obstacle in treatment of leukemia. Therefore, it is urgent to introduce reversing muhidrug resistance into the therapy of leukemia. This article reviewed the research advancements of reversing multidrug resistance in leukemia through RNA interferance technology, which regards the mechanism of multidrug resistance in leukemia as the clue.
