摘要
采用双抗夹心ELISA、APAAP桥联酶标法、MTT比色法检测了34例ITP患儿的血清可溶性白介素2受体(sIL-2R)外周血单个核细胞(PBMC)经PHA刺激后膜白介素2受体(mIL-2R)的表达、T淋巴细胞亚群以及自然杀伤(NK)细胞活性。结果:ITP患儿血清sIL-2R水平升高,mIL-2R的表达和NK活性显著降低;血清sIL-2R水平与NK活性、CD4+/CD8+比值是负相关,与疾病严重程度和疗效密切相关。提示sIL-2R可作为监测病情、观察疗效的敏感指标;在纠正ITP的免疫调节紊乱中应充分考虑到细胞因子受体异常这个环节。
We investigated the serum levels of soluble interleukin-2 receptor (mIL-2R ), the exprexssion of IL-2 receptor (mIL-2R ) on the peripheral blood mononuclear cells (PBMC), T lymphocyte subsets and NK cell activity in 34 0children with ITP and 19 healthy children. Serum levels of sIL-2R were significantly higher, but the mIL-2R expression and NK cell activity were decreased significantly (P <0. 01 ) in children with ITP as compared with healthy controls.Negative correlations were found between serum sIL-2R levels and NK cell activity,and between sIL-2R levels and the ratio of CD4+/CD8+. sIL-2R was associated with disease conditions and the effect of therapy. We concluded that sIL-2R measurement may consider as a sensitive marker of disease condition and response to therapy in patients with ITP. Our investigations indicatd that abnormality of cytokine receptor would be considered in correcting immunoregulation disorder in ITP.
