摘要
菜粉蝶颗粒体病毒京农801株(PrGV-JN801),经蔗糖梯度离心纯化、碳酸钠溶解后,再经Sepharose 2B柱层析分离纯化,纯化的PrGV-JN801包涵体蛋白,在SDS-PAGE上呈1条带。该蛋白含有18种氨基酸,其中天门冬氨酸、谷氨酸和脯氨酸含量较高,酸性氨基酸含量为碱性氨基酸含量的2倍。氨基酸组分值可作为该病毒标准制剂的参考依据。PrGV-JN801包涵体蛋白的分子量为28000道尔顿,紫外吸收特性A_(max)/A_(min)=1.53。其荧光光谱,在pH7时,激发最大值在275nm,其次在235nm,最小值为245nm,发射最大值为304nm;在pH11时,激发光谱为285nm,发射光谱为351nm。
Pieris rapae Granulosis Viruses strain Jing Nong 801 (PrGV-JN 801) were purified by sucrose-gradient centrifugation and then dissolved in sodium carbonate. The dissolved inclusion-body protein which was isolated and purified from the dissolution mixture by using Sepharose 2B column chromatography showed only one protein band by SDS-PAGE. The inclusion-body protein contained 18 kinds ( 195 to 197 moles) of amino acid residues, amoung which Asp, Glu, and Pro were in high percentage. The content of acidic amino acids was twice as much as that of the basic ones. The molecular weight estimated by SDS-PAGE was 2.8×104 daltons.The maximum of UV absorption is 279 nm and the minimum is 250 nm.with a ratio Amax/Amin= 1. 53. The results of fluorescent spectra show that the excition spectra of IBP are 275 nm, 235 nm and 245 nm,emission spectrum is 304 nm at pH 7; while it shifts to 285 nm and 351 nm respectively at pH 11.
