摘要
将构建的三套乙肝病毒基因疫苗(分别编码S蛋白、preS1+preS2+S蛋白及preS2+S蛋白)注射于C57BL/6小鼠胫前肌内。基因疫苗接种3天后,以同样部位注射rhIL-2、rhIL-4、rhIL-6、rhIFN-γ。运用ELISA检测不同时间小鼠血清中乙肝表面抗原(HBsAg)及抗-HBs。结果显示:注射基因疫苗3天内,小鼠体内即可表达HBsAg;两周后,血清中可测到抗-HBs,两月后抗体水平达到高峰,并保持高水平至少两月以上;肌内注射的几种细胞因子均可提高血清中抗-HBs水平,与对照组相比差异有极显著性(P<0.01),其中,rhIL-4、rhIL-6、rhIFN-γ较rhIL-2作用强。提示,细胞因子可作为基因疫苗的佐剂。该研究为增强基因疫苗免疫效果提供了新途径。
Three constructs of HBV genetic vaccines containing S gene,preS1+preS2+S gene and preS2+S gene,respectively,were injected into tibialis anterior muscles of C57BL/6 mice.After 3 days of genetic vaccine inoculation,recombinant cytokines rhIL 2,rhIL 4,rhIL 6 and rhIFN γ were administered at the same sites.ELISA was used to detect the HBsAg and anti HBs in mice sera at various times post gene transfer.The results showed that 3 kinds of plasmids expressed and the HBsAg secreted into serum within 3 days post DNA injection.Anti HBs in sera of mice was tested positive ( P/N >2 1) 2 weeks post plasmids injection,the level of antibody attained the peak 6 weeks later,and kept high titer for at least 2 months.Several cytokines were injected intramuscularly and all could significantly ( P <0 01) promote the production of anti HBs induced by HBV vaccine compared with control group.Among the cytokines, rhIL 4, rhIL 6,rhIFN γ showed more powerful adjuvant effects on HBV genetic vaccine than rhIL 2.The results suggest that cytokines can act as immunological adjuvants for genetic vaccines.The research provides a new way for elevation of immune responses induced by genetic vaccines.
