The Dynamic Expression of TERT,PCNA and Bcl-2 in Epiretinal Membrances in Experimentally Induced Proliferative Vitreretinopathy and their Relativity Study

摘要 目的探讨与细胞增殖相关的端粒酶逆转录酶(TERT)、增殖细胞核抗原(PCNA)和凋亡抑制蛋白Bcl-2在外伤性增殖性玻璃体视网膜病变(PVR)视网膜前膜形成中的动态变化及其相关性.方法用S-P法对外伤性PVR大鼠模型视网膜前膜做端粒酶逆转录酶(TERT)、增殖细胞核抗原(PCNA)、Bcl-2免疫组织化学染色和HE染色,染色结果行图象分析.结果伤后增殖细胞核抗原(PCNA)蛋白表达阳性细胞率和平均光密度值出现先升高后降低的动态变化,14 d组阳性率最高,与7 d组和28 d组比较差异有统计学意义.伤后TERT蛋白和Bcl-2表达阳性细胞率和平均光密度值出现先升高后稍降低的动态变化,14 d组和21 d组阳性率最高,与7 d组比较有显著性差异,TERT、PCNA和Bcl-2蛋白表达之间有显著相关性.结论TERT、Bcl-2参与外伤性PVR视网膜前膜增殖细胞的生长调控,与细胞增殖的动态变化呈高度相关性,为反义核酸技术等基因手段预防或治疗PVR以及治疗时间的选择提供实验依据. Objective To investigate the dynamic change and their correlation of telomerase catalytic sub-unit TERT, proliferating cell nuclear antigen traumatic proliferative vitreoretinopathy. Methods PCNA and Bcl-2 in the formation of epiretinal membranes of S-P technique were applied for immunocytochemical staining of epiretinal membranes of traumatic PVR with TERT, PCNA and Bcl-2 antibody. HE staining was also carried out. The staining results were analyzed with image analysis system. Results The positive rate and average OD of PCNA protein increased at first and then decreased after operation. Compared with 7 days group and 28 days group, the positive rate in 14 days group was highest. The positive rate and average OD of TERT and Bcl-2 also increased at first and then decreased. Compared with 7 days group, the positive rate in 14 days group and 21 days group were higher. There was significant correlation among PCNA, Bcl-2 and TERT protein expression. Gonclusions TERT and Bcl-2 paly roles in the regulation of proliferative cells in epiretinal membranes of traumatic proliferative vitreoretinopathy, and have high correlation with the dynamic change of cell proliferation. It provides experimental basis for preventing or treating PVR by gene technique such as antisense ribozyme and the choice of therapy time.
